Overview of Technology

Nectagen creates open technology platforms that biologists can customize and enhance for the community's benefit. Nectagen develops a critical mass of tools and reagents, supports users' development of new modifications and applications, and ensures open access to the technology for the academic community. Where possible, Nectagen designs its platforms using components that are made available to non-profits on the condition that any modifications and derivatives made by the recipient are in turn made available to other non-profits and Nectagen.

Nectagen's key technology platforms include

nanoCLAMP™ Single Domain Antibody Mimetics. nanoCLAMP (CLostridial Antibody Mimetic Proteins) affinity reagents are recombinant 15 kD antibody mimetic proteins selected for tight, selective and gently reversible binding. The nanoCLAMP scaffold is based on an IgG-like, thermostable carbohydrate binding module from the Clostridium perfringens hyaluronidase (Mu toxin). nanoCLAMPs are 4 nm X 2.5 nm in size, about the same size as a nanobody. We have generated nanoCLAMP libraries with variable loops that are analogous to the complement determining regions of immunoglobulins and used these libraries to isolate nanoCLAMPs recognizing specific immunogens. Our nanoCLAMPs can be used to pull down proteins or protein complexes from whole cell extracts followed by elution with propylene glycol under conditions that preserve activity and protein-protein interactions. Product and Applications Database. Nectagen has organized its applications data in a highly structured, open access database. The database contains detailed protocols, quantitative performance data, and qualitative assessments for different nanoCLAMP-target pairs. The database has the breadth to capture data for almost a dozen classes of applications, including affinity chromatography, ChIP-seq, and standard antibody applications. The database has the depth to provide reference data and details like buffer recipes, starting material strains, and quality control reports for individual lots. Nectagen believes that the best way to build a broad and deep database is to motivate users to contribute data generated in the course of using Nectagen products for their own research. Toward this end, Nectagen provides customers with significant discounts on Nectagen products in exchange for contributing data to the Nectagen community.
